Beschreibung Neurodegenerative diseases disturb all of these functions and dementia is arguably the heaviest medical and personal burden of ageing societies. Movement disorders such as Parkinson’s Disease (PD) and Huntington’s Disease (HD) are further characterized by involuntary motor dysfunction, which are progressively debilitating. Selective degeneration of the corticostriatal-basal-ganglia-thalamocortical loop is prominent in PD and HD and while most research and treatment efforts have been directed towards the basal ganglia, the accessible cortex, located on the surface of the brain has received much less attention.
The underlying mechanisms for PD and HD are far from being well understood. An intersection of molecular changes, structural degeneration and imbalance of communication between individual neurons and across brain regions creates a multilevel complexity, complicating research. Nevertheless, recent years have brought exciting new technologies that enable both systemic gene delivery as well as measurement and manipulation of brain components at unprecedented precision and scale, making the vision of a comprehensive understanding more realistic than ever before.
