Details Eukaryotic genes are generally transcribed as pre-mRNA, in which protein-coding exons are interrupted by non-coding introns that must be excised prior to translation. The molecular machine carrying out the removal of introns and ligation of exons to produce mature mRNA is the spliceosome, a dynamic complex consisting of 5 small nuclear ribonucleoproteins (snRNPs) and over 100 proteins. For each round of splicing, the spliceosome is assembled anew on its pre-mRNA substrate. Formation of the spliceosome’s RNA-based catalytic center occurs during activation, in which the spliceosome’s molecular architecture and composition are drastically altered. In this talk I will present cryo-EM structures of spliceosomes stalled during activation, revealing steps leading to formation of the catalytic center.
