Details | Dipeptide repeat proteins are aberrantly expressed in the brain of ALS/FTD patients due to a mutation in the C9orf72 gene. The neurotoxic mechanism of these dipeptide repeats is associated to their anomalous interaction with cellular proteins, thus interfering with several physiological pathways. We explored a new toxic mechanism of dipeptide repeats that is linked to the inhibition of prolyl isomerases (a class of protein folding chaperones) that disrupt protein homeostasis. |
